Abstet 250mg Tablet

Abstet 250mg Tablet contains its active substance called Abiraterone acetate. It is a CYP17 inhibitor. It is not recommended in paediatric and women. It is used in the treatment of metastasised prostate cancer. It is an anti-testosterone medication used with prednisolone. It is used in cases that have not responded to chemotherapy.

Abstet 250mg is a CYP17 inhibitor also called an adrenal inhibitor used in the treatment of prostate cancer that has metastasised in adults. It acts as an anti-testosterone medication. It is used in the treatment of prostate cancer who have been failed to be treated with chemotherapy. It is not used in children and women.

It is an adrenal inhibitor used in the treatment of castration-resistant, pre-treated with chemotherapy and failed type of prostate cancer. It is used only in adult males. It is not recommended for women and children.

Prostate Cancer : 

Prostate Cancer is also called as prostatic carcinoma. It is a walnut-shaped gland in males which is caused by DNA changes of a normal prostate cell. Symptoms include difficulty in urinating, blood in urine and semen, bone pain, weight loss, and erectile dysfunction

It is an antineoplastic medication and inhibits the multiplication or proliferation of abnormal cells present in the prostate. It is used only in adult males. Abstet is recommended when prostate cancer is

• Castration-resistant
• Chemotherapy with docextal has failed
• Metastasis to other organs

It is used with prednisolone.

Abstet 250mg contains abiraterone acetate is an antineoplastic and a CYP17 inhibitor. Abiraterone acetate is converted into abiraterone which is an androgen inhibitor. It inhibits the enzyme called as 17-alpha-hydroxylase also called CYP17 present in the testis, adrenal glands and prostate. In general, CYP 17, assists in the conversion process that results in the formation of progesterone and androstenedione. Abiraterone acetate can be used in treating androgen dependant prostate cancer. It reduces serum testosterone and progesterone levels. The visible decline in PSA levels is seen with the use of Abiraterone acetate in the treatment of chemo-resistant metastatic prostate cancer. In other words, It treats prostate cancer by inhibiting testosterone formation and decreases serum testosterone levels.

Very Common side effects that occur in more than 1:10 patients: co-infections, nausea, vomiting, loss of appetite, weight loss, eczema, fatigue, hypertension, bleeding disorders, diarrhoea, constipation, rash, thinning of hair, body ache, headache, hypophosphatemia, increase in serum amylase and lipase enzymes

Common side effects that occur 1:10 patients: Hematuria, Pyrexia, Rash, Headache, Urinary frequency, Arrhythmia, Groin pain, Nocturia, Fractures, Chest pain or chest discomfort, Cardiac failure, Lymphopenia, Hypophosphatemia, Hyperglycemia, Increased ALT/AST, Hypokalemia, Joint swelling, Muscle discomfort, Dyspnea, Fatigue, Urinary tract infection, Hematuria,

Uncommon side effects that occur 1:100 patients: Diarrhea, Pyrexia, Hypertriglyceridemia, Groin pain, Constipation, Edema, Hyponatremia, Hot flush, Headache, Insomnia, Increased total bilirubin

• Is Abstet 250mg safe during pregnancy?

No, taking Abstet during pregnancy is not recommended thus inform your physician if you are pregnant or plan to get pregnant

• Can Abstet be taken while breastfeeding?

No, in general, it is suggested that drugs and their metabolites can pass via breastfeeding thus it is suspected that Abstet can also pass via breastfeeding.

• Can Abstet be taken while alcohol consumption?

The interaction is unknown but this medication causes side effects. Thus, consult your physician.

• Can I take a drive or operate heavy machinery while on Abstet?

This medication causes side effects thus caution is used while driving or operating machinery.

• What is the best time to take Abstet?

You should take Abstet on an empty stomach either 1 hr before food or 2 hrs after food intake.

Alcohol

It generally causes side effects thus it is advised not to be taken with alcohol. Kindly consult your doctor.

Driving or operating machinery

It is preferred to drive or operate heavy machinery with caution.

Pregnancy

It is highly unsafe and not recommended during pregnancy. Kindly consult your doctor

Breast Feeding

It is not safe during breastfeeding it may be toxic to the baby. Kindly consult your doctor.

Kidney Problem

No dose adjustment is required for mild to moderate kidney disease patients but for severe kidney disease patients dose adjustment with caution is required. Kindly consult your doctor before usage.

Liver Disease

Dose adjustment may be required with caution for patients with liver disease. Kindly consult your doctor before usage.

• It Should be taken orally.
• It must be taken at the same time once a day as prescribed by your physician.
• It should be taken without food either 1 hr before food or 2 hrs after food intake
•  It must be swallowed as a whole.
• Don’t chew or crush the tablet.
• It must be taken with prednisolone.
• After therapy has been initiated, do not take more or less than the prescribed dose or stop taking it without your physicians’ approval. Skipping or changing the Abstet dose without approval may cause adverse effects making the condition more difficult to treat, or worsen side effects.

If you are pregnant or breastfeeding inform your physician as they are not advisable.

• Allergic reactions - It can be mild hypersensitivity and angioedema to severe like Stevens-Johnson Syndrome and toxic epidermal necrolysis.
• Electrolyte imbalance – causes hypokalemia/ hypertension/ edema. monitor function before and during treatment
• Adrenocortical insufficiency
• Hepatotoxicity– monitor function before and during treatment
• Increase risk of bone fracture
• Renal toxicity – minimal to none
• Pregnancy – causes embryo-fetal toxicity
• Male infertility
• Breastfeeding – causes side effects similar to adults
• Alcohol intake or Driving or operating heavy machinery – Abstet causes side effects thus caution is required. consult your physician

It should be taken orally as prescribed dosage at the specific time regularly without food either 1 hr before food or 2 hrs after food intake to avoid adverse effects. This tablet is always taken with prednisolone.

Drug-Drug Interaction

Serum concentration is decreased by Dexamethasone and other steroids

Serum concentration is increased by Carbamazepine, St. John's Wort, Omeprazole

Drug-Food Interaction

It should be taken either 1 hr before or 2 hrs after food, if doubts arise kindly refer to your physician.  Abiraterone acetate generally causes side effects thus it is advised not to be taken with alcohol, spicy foods, caffeinated beverages.

Drug-Disease Interaction

• Electrolyte disorders – used with caution in hypokalemia causing diseases. Consult a physician before use
• Liver disorders – used with caution in severe liver diseases. Consult a physician before use
• Kidney disorders – used with caution in severe kidney diseases. Consult a physician before use
• Cardiac disorders – used with caution in heart failure and MI. Consult a physician before use
• Respiratory disorders – used with caution in respiratory distress. Consult a physician before use
• Blood disorders – used with caution in haemophilia or anaemia. Consult a physician before use
• Adrenal disorder – used with caution in adrenocortical dysfunction. Consult a physician before use

Overdosage of abiraterone acetate has less literature kindly consult your doctor.

You can take the missed dosage if you notice within 12 hours. If it is more than 12 hours, then skip the dosage and take the next doses as usual. Don’t take a double dose for a forgotten dose.
