Karplat 450mg Infusion

Karplat 450mg Infusion contains Carboplatin 450mg it’s an antineoplastic medication and alkylating agent. It is a platinum analogue. It is used in the treatment of ovarian cancer. It is used as an orphan drug in the advanced stages of cervical cancer, endometrial cancer, esophageal cancer, head and neck cancer, melanoma, non-small cell lung cancer, sarcoma, small cell lung cancer, testicular cancer, and thymoma. It is also used as an orphan drug in metastatic breast cancer and neuroendocrine carcinoma of the skin.

Karplat 450mg Infusion is used in the treatment of advanced ovarian cancer. It is not used in neonates. It is also used as an orphan drug in several cancers and leukemia.

Carboplatin is an alkylating medication. It causes intra- and inter-strand crosslinks between DNA bases causing break in DNA strand thus apoptosis is induced resulting in cell death of abnormal DNA containing cancer cells.

Carboplatin is an antineoplastic and alkylating medication. It causes DNA base pairs to break inducing apoptosis of these abnormal cancer cells.

Absolute contraindication:

• if you are allergic to Karplat 450mg (Carboplatin 450mg) Infusion or its constituents
• if you are pregnant
• if you are breastfeeding

Relative contraindication:

• if you are vaccinating
• if you have a kidney disorder
• if you have a neurological disorder
• if you have a lung disorder
• if you have a heart disorder
• if you have a liver disorder
• if you are a smoking/tobacco user

  Common side effects that occur 1:10 patients: Peripheral neuropathy, Anemia, Nausea, Fatigue, Diarrhea, Vomiting, Abdominal pain, Constipation, Thrombocytopenia, Fever, Anorexia, Leukopenia, Dyspnea, Cough

Uncommon side effects that occur 1:100 patients:Edema, Neutropenia, Pharyngolaryngeal dysesthesia, Anaphylactic-like reaction, Pulmonary fibrosis,hypokalemia

Rare side effects that occur 1:1,000 patients: Pulmonary fibrosis, Posterior leukoencephalopathy syndrome, Loss of deep tendon reflexes, dysarthria, Lhermitte’s sign, cranial nerve palsies, fasciculations, convulsion, RPLS, Deafness, Laryngospasm, colitis, metabolic acidosis, ileus, intestinal obstruction, pancreatitis; veno-occlusive disease of liver, perisinusoidal fibrosis, Hemolytic uremic syndrome, Acute tubular necrosis, acute interstitial nephritis, acute renal failure, Pulmonary fibrosis, interstitial lung diseases, Rhabdomyolysis, Septic shock

No, taking Karplat 450mg Infusion during pregnancy is not recommended thus inform your physician if you are pregnant or plan to get pregnant

This Infusion is used only in the presence of physician; Do not shake medicine containing vial while diluting or infusing; It is compatible with D5W; Do not use NS or chloride containing solutions; It is infused into 250-500ml of D5W and administered as slow infusion; Refrigerate unopened or opened 8?C

After therapy has been initiated, do not take more or less of this drug than prescribed or stop taking it without your physicians’ approval. Skipping or changing your dose without approval may cause adverse effects making the condition more difficult to treat, or worsen side effects.

If you are pregnant or breastfeeding inform your physician as they are not advisable.

This Infusion is not recommended in children less than 2 years of age. If used to be done so only with caution.

This Infusion has only been used in few patients 65 years or older. Dosage and usage are similar to adults. Discuss with your physician before usage.

• Allergic reactions - It can be mild hypersensitivity and angioedema to severe like Stevens-Johnson Syndrome and toxic epidermal necrolysis.
• Renal toxicity – impaired renal clearance
• Neurotoxicity – causes tremors
• Avoid cold food/air for next 4 days
• Pulmonary toxicity – pulmonary fibrosis
• Gastrointestinal effects -
• Cardiotoxic – MI/CHF/AF/bradycardia/prolonged QT syndrome
• Electrolyte imbalance – hypokalemia
• Reversible posterior leukoencephalopathy syndrome
• Rhabdomyolysis
• Hepatotoxic – impaired liver enzymes
• Pregnancy – causes embryo-fetal toxicity
• Breast feeding – causes side effects similar to adults
• Alcohol intake or Driving or operating heavy machinery – Karplat 450mg Infusion causes side effects thus caution is required. consult your physician

This Infusion is was given intravenously in the presence of a physician. It must be kept at 8°C. Do not store after a single-use.

Drug-Drug Interaction

Severity of adverse effects is increased by Allopurinol, Beclomethasone

Severity of adverse effects is increased by Carboplatin combined with Arsenic trioxide, Doxorubicin

Severity of nephrotoxicity is increased byAceclofenac, Acyclovir, Etoposide, Mefenamic acid

Severity of ototoxicity and nephrotoxicity is increased by Capreomycin, Dihydrostreptomycin, Gentamicin

Severity of bleeding is increased by Abciximab, Antithrombin Alfa

Drug-Food Interaction

This medication can be taken with any type of food, if doubts arise kindly refer to your physician. It generally causes side effects thus it is advised not to be taken with alcohol. It is preferred to drive or operate heavy machinery with caution. Do not smoke while taking this medication. Do not use tobacco while taking this medication.

Drug-Disease Interaction

• Cardiac disorders – used with caution in QTc prolongation. Consult a physician before use
• Hematological disorders – used with caution in bleeding disorders like thrombosis. Consult a physician before use
• Bone disorders – used with caution in osteonecrosis. Consult a physician before use.
• Liver disorders – used with caution in severe liver diseases. Consult a physician before use
• Kidney disorders – used with caution in severe kidney diseases. Consult a physician before use
• Respiratory disorders – used with caution in respiratory distress and interstitial lung disorder. Consult a physician before use
