Spegra Tablet contains Dolutegravir 50mg, Emtricitabine 200mg and Tenofovir Alafenamide 25mg. Spegra tablet (Dolutegravir (50mg) + Emtricitabine (200mg) + Tenofovir Alafenamide (25mg)) is an antiviral medicine used in the treatment of Human Immunodeficiency Virus (HIV) – 1 infection. Dolutegravir is an integrase strand transfer inhibitor. Emtricitabine and Tenofovir Alafenamide is a nucleoside reverse transcriptase inhibitor. Spegra tablet works by reducing the viral load of HIV in your body. Spegra tablet improves your immune system and reduces the risk of conditions associated with HIV infection. 

Spegra tablet (Dolutegravir (50mg) + Emtricitabine (200mg) + Tenofovir Alafenamide (25mg)) tablet is used to treat adults and children above the weight class 40kg only who are infected by HIV-1. Spegra tablet is not indicated for its use as pre-exposure prophylaxis.

Spegra tablet (Dolutegravir (50mg) + Emtricitabine (200mg) + Tenofovir Alafenamide (25mg)) tablet is used to help control HIV infection. Spegra tablet helps to decrease the amount of HIV in your body thus your immunity works better by reducing your chance of getting HIV complications like co-infections and other cancers. Spegra tablet is not a cure for HIV infection, it improves immunity that results in longevity of life. It is recommended for use in children from 40 kgs and in adults.

Spegra tablet (Dolutegravir (50mg) + Emtricitabine (200mg) + Tenofovir Alafenamide (25mg)) tablet is indicated in the treatment of HIV infection that causes AIDS in humans. Spegra tablet boosts the immunity of the affected hosts. Spegra tablet is indicated in adults and children above 40kgs.

Spegra Tablet contains Dolutegravir 50mg, Emtricitabine 200mg and Tenofovir Alafenamide 25mg. Dolutegravir inhibits strand transfer activity of integrase in DNA thus blocks replication of HV-1. Emtricitabine is an analogue of cytidine that is phosphorylated to its active form Emtricibine 5 triphosphate that inhibits the activity of HIV-1 reverse transcriptase and incorporates themselves into the DNA thus results in termination of the replication process. Tenofovir Alafenamide enters hepatocytes where it is converted to tenofovir by hydrolysis. Hepatocyte tenofovir is then phosphorylated into tenofovir-diphosphate that incorporates themselves into the DNA strand thus stops the replication of viral particles. Thus, the active HIV molecules become inactive and further reproduction and multiplication process is stopped.

Spegra tablet (Dolutegravir (50mg) + Emtricitabine (200mg) + Tenofovir Alafenamide (25mg)) tablet is an integrase inhibitor and nucleoside reverse transcriptase inhibitor. Dolutegravir inhibits the transfer of integrase in DNA and Emtricitabine and tenofovir incorporates themselves into the DNA thus stops the replication of viral particles. Thereby preventing the formation of mature virus particles into infectious virions that spreads the infection.

Absolute contraindication:

• if you are allergic to Spegra (Dolutegravir (50mg) + Emtricitabine (200mg) + Tenofovir Alafenamide (25mg)) tablet and its contents
• if you are pregnant
• if you are diabetic

Relative contraindication:

• if you have a liver disorder like hepatitis B or C or Child-Pugh disease
• if you have blood disorders
• if you have renal diseases

Very common side effects that occur in more than 1:10 patients: Diarrhea, headache, nausea

Common side effects that occur 1:10 patients: Rash, pruritus, vomiting, Flatulence, Abdominal pain, Anemia, Allergic reaction, Impaired concentration, Lipodystrophy, Metabolic acidosis, Leukopenia, Thrombocytopenia, Weakness, insomnia, abnormal dreams, depression dizziness, tiredness, changes in LFT and increased creatine kinase

Uncommon side effects that occur 1:100 patients: lactic acidosis, hepatitis B flare up, suicidal thoughts and behaviours, joint and muscle pain

Rare side effects that occur 1:1000 patients: osteonecrosis

Spegra tablet (Dolutegravir (50mg) + Emtricitabine (200mg) + Tenofovir Alafenamide (25mg)) has only been used in few patients 65 years or older. Dosage and usage are similar to adults. Discuss with your physician before usage.

• Blood disorders Hemophilia, hemolytic anemia
• Skin reactions - It can be mild to severe like Stevens-Johnson Syndrome and toxic epidermal necrolysis.
• Musculoskeletal problems - Osteonecrosis that causes joint stiffness and pain in larger joints like shoulder, hip, and knee
• Accumulation or Redistribution of body fat or
• immune reconstitution syndrome.

In general, if you take once a day dosage, the missed single dose of Spegra tablet can be taken within the next 12 hrs, you can take it. If it is more than 12 hrs, skip the dose and take the next dose at the recommended time. Don’t take a double dose for a forgotten dose.
